<description>Title: NutriTrace: A Living DNA Recorder for Fermentation Description: In a fermentation tank, cells can experience local stress even while fixed probes report normal conditions. Once conditions recover, the event can drop out of the data. NutriTrace is an engineered yeast-based recorder designed to preserve these otherwise overlooked stress histories in DNA. By connecting the recording system to suitable sensing modules, the platform could be adapted to different abnormal conditions. In this project, we selected glucose and oxygen limitation as two representative conditions for development and testing. Our design connects stress-responsive promoters to Cre and FLP recombinases, using mutant recognition sites to favour stable DNA changes. Intron splicing and T2A-mediated ribosomal skipping support a compact fluorescent readout, while alginate-chitosan microbeads help retain the engineered cells and allow small molecules to diffuse in. This video presents the development and testing of our sensing, DNA-writing, fluorescence-readout and encapsulation modules, alongside computational modelling and the development of FLP-Var, an optimised FLP coding sequence. Feedback from industry and the public informed our design priorities, while educational activities engaged different audiences. Through these two initial applications, we are developing a broader approach to recording what cells experienced, even after their surroundings return to normal. Wiki | Official Page</description>
