<description>PerioGuard — an engineered probiotic and a porous coating, designed to bring regenerative signals closer to the tissue that needs them. This overview follows the project's two-part design. The problem. Periodontitis does more than inflame the gums: it damages the tissues and the alveolar bone that hold teeth in place. Stopping the damage matters, but rebuilding what was lost is the harder problem. The design. A single molecule, 2FA, takes on two separate roles. Living module. Inside engineered E. coli Nissle 1917 (EcN), 2FA is converted to xanthine. A HucR-responsive circuit then controls expression of CsgA–AMG, and CsgA helps bring AMG outside the cell, where it can act on periodontal tissue., Material module. Separately, 2FA and CA self-assemble into PNAC (porous nucleoside-based adsorptive coating), a hydrogen-bonded framework that associates with the EcN surface, strengthening the material–bacterium association and reducing early phagocytic clearance., Where they meet. PNAC coats engineered EcN, combining protein delivery with a protective material interface — a living-material system designed to act locally at the periodontal site.
