Against the background of the fast-growing global pet oral care market, periodontal inflammation has become a severe threat to companion animals. Pathogens such as Fusobacterium nucleatum (Fn) and Porphyromonas gingivalis (Pg) secrete virulence factors that activate inflammatory pathways and cause continuous damage to periodontal tissues. Conventional clinical treatments, dominated by antibiotics and synthetic chemical agents, have obvious limitations: long-term use of antibiotics induces pathogen drug resistance, while synthetic chemicals may leave harmful residues. To solve these problems, we developed a gentle biological regulation strategy with three core modules: 1.Precise pathogen clearance: Engineered M13 phages based on phage display technology carry targeting peptides to specifically recognize Fusobacterium nucleatum, and deliver metronidazole for targeted elimination of pathogenic bacteria. 2.Gingival microenvironment repair: Engineered E. coli Nissle 1917 (EcN) probiotics serve as a "repair support team", secreting the IL-10 repair factor at gingival inflammation sites with high reactive oxygen species (ROS) levels to restore gingival health. 3.Sustained targeted delivery: Smart hydrogel microspheres stably encapsulate the engineered bacteria and phages, realizing targeted retention and long-lasting release of active components in the oral cavity.
