Title: Dual EcN Probiotic Synergistic Therapy for the Rare Disease Hyperlysinemia Description: Hyperlysinemia is a rare amino acid metabolic disorder. Patients cannot break down lysine in their bodies and must maintain a lifelong low-protein diet. This condition causes severe complications including brain developmental delay and epilepsy, trapping patients in a lifelong “dietary prison”. Our project presents dual engineered EcN probiotics for synergistic therapy. We build lysine degradation pathways inside EcN strains to consume excess lysine. By applying the SpyTag/SpyCatcher system, we physically couple these two EcN strains to construct metabolic tunnels, mitigating low reaction efficiency and secondary toxicity from intermediate diffusion. A kill-switch is also integrated into our engineered strains to manage biosafety risks from bacterial leakage. Our therapy aims to free hyperlysinemia patients from strict lifelong dietary restrictions and deliver a promising new treatment option. Wiki | Official Page
