Anaerobic pathogens such as Fusobacterium nucleatum proliferate in subgingival biofilms, causing gingival inflammation and progressively destroying periodontal tissues and alveolar bone. Conventional treatments rely on broad-spectrum antibiotics and synthetic chemicals, raising concerns about drug resistance and harmful residues. We designed a dual-payload, pH-responsive hydrogel delivery system for local treatment of pet periodontitis. An engineered, non-pathogenic and strongly colonizing probiotic EcN delivers the anti-inflammatory cytokine IL-10, while a metronidazole-loaded chimeric M13 phage specifically targeting F. nucleatum provides antibacterial activity. Both are separately encapsulated in a sodium alginate–oligofructose hydrogel with a pH-responsive coating, enabling controlled release within the inflamed microenvironment. For Human Practices, questionnaire surveys and multiple stakeholder interviews repositioned our product as post-scaling maintenance and daily prevention, guiding the optimization of the gel formulation and delivery method. We also carried out campus and public outreach events, fostering two-way dialogue between synthetic biology and society. In future work, we will incorporate CRISPR-Cas9-based biosafety designs to further enhance the safety and controllability of the engineered live bacteria.
