Title: G-SCOUT (Gut-specific Synthetic Cellular Optimized Unit for Tumor-detection) Description: Colorectal cancer (CRC) is highly treatable if detected early, yet existing screening methods suffer from low compliance. G-SCOUT aims to solve this by engineering a non-invasive, at-home early screening tool using a probiotic E. coli Nissle 1917 chassis. Our system features an OR-gate genetic circuit that detects Fusobacterium nucleatum (enriched up to 132-fold in early-stage CRC) via ethanolamine and co-aggregation signals. Once triggered, the bacteria produce a visible pink pigment (spisPink) for naked-eye stool interpretation, intentionally avoiding the natural yellow-green spectrum. To ensure safety, we engineered an oxygen-responsive self-lysis switch, ensuring the bacteria die upon excretion. Guided by our Integrated Human Practices (iHP) toolkit—including the CARE framework and ethical checklists—we iterated our design through deep stakeholder dialogue. G-SCOUT connects synthetic biology with practical public health, aiming to provide an accessible, highly sensitive, and responsible screening solution. Wiki | Official Page
