<description>Title: PotyGuard+- A Bodyguard for your vegetable yard Description: In this Video, we, iGEM Team Bonn 2026, introduce PotyGuard+. Potyviruses are one of the most devastating and widespread plant diseases, threatening food security around the globe. Current strategies for fighting viral infections in plants are very limited. Our goal is to engineer a mechanism that combats viral spread at the cellular level. We introduce Potyguard+, a protease-dependent modular toolbox that uses the activity of the conserved potyviral NIa-type protease as a signal for an antiviral response. After infection, the NIa-type protease activates our system by cleaving off inhibitory cages, allowing for the reconstitution of Cre recombinase via intein trans splicing. Cre recombinase then cuts the plant's DNA at predetermined loxP sites, activating an antiviral response by enabling the expression of the non-specific RNase Barnase. This modular system, which separates sensing of the protease activity from the response via Cre recombinase, can in turn easily be modified to the desired severity of the antiviral response without the need for a redesign.
