Title: Dual-Response EcN: An Adjunct Therapy for Pseudomonas Infections in cystic fibrosis Patients Description: This video introduces our synthetic biology therapy targeting chronic Pseudomonas aeruginosa biofilm infections in cystic fibrosis patients. These patients often suffer from recurrent drug-resistant lung infections, and effective targeted clinical treatments remain scarce. We engineered an EcN strain with a coupled Fur and RhlR sensing circuit for dual-specific recognition. The Fur system is de-repressed only in iron-deprived mucus at infection sites to achieve spatial localization, while RhlR detects AHL quorum-sensing signals secreted by Pseudomonas aeruginosa. Expression of alginate lyase and Lysis E is triggered exclusively when both low iron and high AHL signals are present. Lysis E lyses EcN cells to release alginate lyase, which breaks down the biofilm matrix of Pseudomonas aeruginosa and improves antibiotic performance. Integrating environmental and pathogen sensing, this design enables targeted localization and in situ biofilm clearance for chronic respiratory infections. Wiki | Official Page
