https://video.igem.org/videos/watch/84926d28-c603-472b-b728-94f8c98213a6 Title: Engineering a Yeast Biosensor for Influenza A HA Recognition and Environmental Risk Screening Description: We aim to engineer Saccharomyces cerevisiae to detect influenza A hemagglutinin (HA), targeting future public-space environmental monitoring. Inspired by antigen-gated GPCRs, our design connects HA recognition to yeast MAPK signaling and FUS1-driven reporters. Through iterative Design–Build–Test–Learn cycles, we developed four modules: reporters, FAR1/SST2 chassis modifications, humanized Gpa1, and antigen gating. Reporter and chassis-engineering modules were constructed and functionally characterized. Computational modeling prioritized nanobody R1a-B6 and proposed interaction-optimization hypotheses. We tested hM1Dq- and Ste2-based gating designs. Ste2-based constructs retained α-factor-responsive fluorescence, but neither design demonstrated HA-dependent activation, highlighting gating and coupling challenges. We also developed a temperature-controlled optical-readout prototype and incorporated stakeholder feedback to refine application requirements. Our contributions include reusable genetic parts, characterized engineering resources, and documented design iterations. HA-dependent sensing and air-sampling integration remain unvalidated; this work provides an experimental foundation rather than a deployment-ready detector.
