Title: preCISe: a modular tool for intracellular delivery using contractile injection systems Description: Protein therapeutics offer unique advantages in terms of versatility and specificity, making them promising candidates for treating a wide range of diseases. However, their inability to efficiently cross cell membranes generally limits their access to intracellular targets. preCISe addresses this challenge with a platform for target-specific protein delivery based on extracellular contractile injection systems (eCISs), bacterial nanosyringes that inject cargo directly into the cytosol. The platform comprises a modular eCIS design that can be retargeted to new receptors by exchanging their binding domains, preCISion reporter cell lines that quantify intracellular delivery, and our deCISion software which finds cell-specific receptors and binders and returns ready-to-order DNA sequences. Because receptors unique to one cell type are rare, we initiated a dual-targeting strategy: the cargo is split between two eCISs addressing two receptors and reassembles only in cells displaying both. Our software OpenSplitProtein ranks split sites that preserve folding and spontaneous reassembly. Wiki | Official Page
