<description>Title: ZipTide: designing short peptides against zonulin-driven loss of the intestinal barrier Description: The intestinal barrier is one cell thick, and zonulin loosens the tight junctions that hold it shut. No approved antagonist exists. ZipTide designs short peptides against that signal. We collapsed the 25.6-billion-sequence space of eight-residue peptides onto 2,220,075 compositions without discarding any, ranked them by physicochemical distance to larazotide, screened 1,500 through three published predictors, and carried twenty to structure prediction and docking. Three were chemically synthesised and deposited in the Registry alongside the control. Caco-2 cells challenged with gliadin were read as zonulin release by ELISA, against a criterion written before any data existed. The assay fired: gliadin raised zonulin about fivefold and larazotide brought it down. Neither candidate tested reached the bar; the third arrived as an empty vial. Twenty-three stakeholder conversations reshaped the project's scope, read-out and language, and thirteen of them changed it. This video says plainly what the project is not.
