<description>Title: Fridge Guardian — A Photocontrolled AND-Gate Biosensor for Viable Listeria monocytogenes Description: Can we detect living Listeria monocytogenes before contaminated food reaches the table? This UM-Macau iGEM 2026 team presentation introduces Fridge Guardian, a portable smartphone-based biosensor built around a molecular AND gate. In the wet lab, we develop four modular strategies that convert LLO-triggered membrane pore formation and light-controlled split-sfGFP complementation into fluorescent signals. In the dry lab, we integrate mathematical modeling, a smartphone fluorescence detector, and an image-analysis pipeline to translate optical signals into quantitative estimates. Through Integrated Human Practices, public survey, consultations with stakeholders, outreach with other teams, regulatory analysis, and community education, these directly shaped the project’s design and validation. By linking LLO-triggered membrane pores with photo-controlled fluorescence, Fridge Guardian aims to make rapid, specific testing of viable Listeria more accessible in real food-safety settings.
