Title: Capture-Striking Back at Pseudomonas aeruginosa with Dual-Action Engineered E. coli Description: Hospital-acquired infections are a silent global health crisis, and at the forefront of this battle is Pseudomonas aeruginosa—a notorious pathogen that builds highly resistant biofilms and releases deadly toxins. But what if we could intercept its communication before it strikes? Meet our iGEM 2026 project - Capture! We have engineered a revolutionary dual-action E. coli system designed to outsmart P. aeruginosa, serving as both an early-warning diagnostic tool and an emergency first-line treatment. Here is how our engineered bacteria tackle the problem: 1. Passive & Low-Cost Diagnosis By detecting the AHL molecules (quorum-sensing) of P. aeruginosa, our engineered E. coli triggers a visible fluorescent signal. This allows medical professionals to identify infections early, accurately, and at a fraction of the cost of traditional methods. 2. Emergency First-Line Intervention Diagnosis is just the beginning. Our system features a novel reverse transcription mechanism that produces specific single strand DNA aptamers on demand. These aptamers act as biological interceptors, blocking the pathogen’s quorum-sensing communication. The result? We effectively prevent biofilm formation and stop the release of toxins. By providing rapid diagnosis and immediate intervention, our system strips the pathogen of its defenses, significantly improving the effectiveness of subsequent and traditional antibiotic treatments. We are not just fighting the bacteria; we are disarming them. Wiki | Official Page
