<description>Title: CloBiT: Expanding the Toolkit for Clostridium butyricum Description: Clostridium butyricum is a highly promising non-model organism with applications ranging from probiotics to bioenergy, but its potential is limited by a lack of characterized genetic tools. Our project, CloBiT, bridges this gap by expanding the genetic toolkit for this anaerobic chassis. We characterized baseline growth under stress and validated conjugation protocols. To address limited gene regulation, we developed a computational pipeline utilizing a 3-model ensemble (Evo v2, CNN v2, ProkBERT) to mine and rank promoter candidates. We screened these computationally predicted promoters experimentally in E. coli to assess their expression strength. Our Human Practices work explored the regulatory and biosafety landscape for live biotherapeutics, revealing critical insights on containment strategies and guiding our design. Furthermore, our education programs engaged over 200 people across diverse communities. By organising events focused on mutual learning and open dialogue on synthetic biology and gut health, we turned passive learning into active science communication. By providing a documented, versatile promoter toolkit and fostering public dialogue, CloBiT empowers future researchers to explore non-model organisms and drive innovation in synthetic biology.
