Title: THULIR Description: Seeds are the lifeline of plants, facilitating their propagation and conservation. Orthodox seeds withstand conventional seed banking methods, remain viable for extended periods. In contrast, recalcitrant seeds are highly sensitive to dehydration and freezing, making their long-term preservation challenging, endangering the conservation of ecologically important plant species. This is due to high expression of Late Embryogenesis-Abundant (LEA) proteins in orthodox seeds where it protects the seeds from stress. THULIR provides a solution by engineering the LEA domains with metal ion binding motif and a cell-penetrating peptide, that facilitates their delivery into the embryonic axis of the Artocarpus heterophyllus seeds. LEA proteins are engineered with pET (blank) vector and expressed in E. coli BL21, purified through His-tag/Ni-NTA column chromatography. Once delivered, the engineered LEA proteins enhance the cryo-stress tolerance and improve their survival without modifying the plant genome, facilitating a sustainable strategy for long-term cryopreservation. Our custom designed software, StressPep would help the researchers to find a suitable protein for stress related studies in future, and also our hardware, CryoAxis, an automated system supports controlled cooling and solution handling, during the cryopreservation of embryonic axes or seeds of recalcitrant plant species. We duly acknowledge the contribution of Scientists, Research scholars, Farmers and Institutions for the successful completion of our project. Wiki | Official Page
