        <description>Title: Pine Protector Description: This project employs the naturally endophytic bacterium Bacillus subtilis— a resident symbiont of pine wood — as the chassis organism. Using synthetic biology approaches, the engineered strain is designed to heterologously express alkaline serine protease (BLG) for targeted nematocidal activity against the pine wood nematode. Concurrently, it produces the coleopteran-specific Bacillus thuringiensis toxin Cry3A to suppress the vector insect (Japanese pine sawyer beetle). A self elimination system based on the KillerRed protein is incorporated, which is activated upon light exposure once the engineered bacteria leave the internal pine environment, thereby triggering programmed cell suicide to ensure biocontainment.
