Title: SweetCycle: Enzyme Engineering of Steviol Glycosyltransferases for Enhanced Rebaudioside M Production Description: Reducing sugar without compromising taste demands better sweeteners. Rebaudioside M (Reb M) offers a clean, sucrose-like taste, but its low natural abundance and costly purification limit wider use. SweetCycle combines glycosyltransferase engineering with Stevia hairy-root screening to address this challenge. We constructed eight variants of SrUGT91D2 and SrUGT76G1 and evaluated them through tobacco transient expression followed by screening in Stevia rebaudiana hairy roots. In this preliminary screen, SrUGT76G1 T284I showed approximately 42% higher estimated Reb M content than wild type, identifying a promising candidate for further validation. Guided by stakeholder engagement and techno-economic modeling, we envision applying these candidates to enzymatic conversion and using hairy-root screening to support future crop improvement. Our work connects enzyme design with evaluation in the source plant, laying the groundwork for more accessible, better-tasting stevia ingredients. Wiki | Official Page
