Title: NariViva- Engineered Probiotics Delivering Naringenin to Restore Vaginal Health Description: Genitourinary Syndrome of Menopause (GSM) is a chronic condition associated with declining estrogen levels, affecting quality of life in postmenopausal women. NariViva aims to develop a probiotic-based biomedical platform for localized, pH-responsive naringenin production as a potential strategy for GSM management. We designed a genetic circuit integrating a pH-responsive regulatory module with a naringenin biosynthetic pathway comprising tyrosine ammonia-lyase (TAL), 4-coumarate-CoA ligase (4CL), chalcone synthase (CHS), and chalcone isomerase (CHI). Using Escherichia coli as an initial chassis, we characterized pH-responsive gene expression and investigated a naringenin-responsive regulatory module. Western blot and HPLC analysis showed increased CHI abundance with rising pH in the circuit without negative feedback, whereas the negative-feedback design helped stabilize CHI abundance across the tested pH range. These findings support the feasibility of combining environmental sensing with feedback regulation. Further product-level validation, chassis optimization, and biosafety evaluation are needed before future development in vaginally compatible probiotics. Wiki | Official Page
