<description>Title: Powering Synthetic Cells: Education, Energy, and Engineering Description: Synthetic cells are cell-free protein synthesis reaction encapsulated in lipid membranes, offering a controllable, non-living platform for studying cellular processes such as genetic information processing, and energy metabolism. We built an undergraduate lab course at Cal Poly around them, and student feedback showed us how to make the techniques more reproducible. In response, we designed a 3D-printed emulsion device that standardizes preparation across multiple tubes and a “tip wipe” method that bypasses oil removal. To further optimize the platform, we screened 16 uncharacterized class III PPK2 enzymes as ATP-regenerating parts that address the energy limits of cell-free reactions, titrating enzyme, magnesium, and DNA concentrations to find their ideal conditions. To measure enzyme activity reliably, we developed a luciferase-based ATP assay with effective quenching methods and outputs corrected for substrate and quench inhibition. Our workflow gives any team a way to test new parts while keeping the work safe and accessible and engaging stakeholders. Our goal is to inspire other iGEM teams, industry professionals, and institutions to continue engaging with non-experts for the benefit of the broader synthetic biology community.
