Title: CirCure: A Multi-Epitope Circular RNA Vaccine Platform for Triple-Negative Breast Cancer Description: Triple-negative breast cancer (TNBC) is hard to treat: its tumors are heterogeneous, recur often, and escape single-target therapies. CirCure is a modular circular RNA (circRNA) vaccine platform that broadens immune recognition beyond a single TNBC-associated antigen. A computational pipeline screens antigen fragments, or epitopes, for MHC-I binding, immunogenicity, and structural compatibility, prioritizing candidates with strong potential to activate CD8+ "killer" T cells. The selected epitopes are encoded into a single engineered circRNA. Inside dendritic cells, the circRNA acts as a durable instruction set, supporting antigen production and presentation so CD8+ T cells can recognize and attack tumor cells with different antigen profiles. We experimentally validate circRNA circularization, antigen expression, dendritic cell maturation, and T-cell-mediated tumor cell killing. Input from clinicians and other stakeholders guides our iterative design. CirCure offers a scalable, "plug-and-play" framework for RNA vaccines against cancers in which tumor diversity limits single-target treatments. Wiki | Official Page
