<description>Title: RATAtouille, Multi-Layer Genetic Circuits for Dynamic Metabolic Control Description: In microbial production, performance is decided less by which genes are present than by when they switch on. Static control fixes expression once and holds it for the whole culture, while the cell moves through growth and production. Four costs follow: resources skew to the product and growth slows, intermediates accumulate and inhibit, flux leaks at branch points, and closing a branch collapses growth. RATAtouille, a Reprogrammable Autoregulatory Transcriptional Architecture, uses what the cell already knows. Input modules reading precursor level, population density and external signals share one output format, so physiological context is read from several angles. Hierarchical logic turns that combination into a single expression state, and the delay through each layer divides the pathway in time. Our first application is lutein, where one promiscuous cyclase feeds both product and byproduct and cannot be deleted. We change only when it is expressed, so substrate ratio decides the flux.
