<description>Title: dogmaOS: Engineering Translation Rhythm for Improved Soluble Protein Expression Description: Soluble expression of heterologous proteins in Escherichia coli remains challenging, as efficient translation does not guarantee proper folding. We developed dogmaOS, a platform that aims to improve protein folding by engineering translation rhythm and the cellular folding environment. Our approach integrates computational modeling, synonymous codon redesign, and host tRNA pool engineering. Using Four-Leaf Clover RT-qPCR and ribosome profiling, we quantified changes in tRNA abundance and investigated their effects on translation elongation. To address disulfide-bond formation, we further incorporated signal peptide-mediated periplasmic targeting. Combining these strategies significantly improved the soluble-to-insoluble ratio of TolB-G-CSF compared with its commercially optimized counterpart. Beyond the laboratory, stakeholder engagement and inclusive educational initiatives connected our research with real-world needs. Together, dogmaOS establishes a measurement-guided framework integrating sequence design, host engineering, and protein targeting, offering new possibilities for recombinant protein production.
