<description>Title: TEMPO: Timed Expression and Suicide Module with Programmable Oscillator-Coupled Binary Counter Description: Biological functions depend not only on what cells do, but also on when they act. Many existing genetic timing strategies couple timekeeping directly to a specific downstream function, so adapting them to a new task often requires redesigning the timing circuit itself. TEMPO instead separates timekeeping from execution, providing a modular internal clock that can connect to different downstream programs. It integrates three modules: an oscillator-based CLOCK that produces periodic internal signals, a reversible MEMORY that records successive signals as binary DNA states, and an ACTUATOR that links selected counts to specific outputs. Binary encoding enables multiple stages to be represented without adding a complete regulatory layer for every new step. By tuning the clock period, target count, and output gene, the architecture provides a general framework for staged biomanufacturing, sequential research perturbations, timed therapeutic programs, and programmed elimination for biocontainment.
