ECUST: ABM Simulation 2: Light-Off Control of E. coli Mucus Penetration (2026)

This video presents a light-off control simulation of engineered Escherichia coli migration across a fluid–mucus interface using our agent-based model (ABM). The animated x–y occupancy heatmap visualizes the cumulative spatial distribution of bacterial trajectories over time, with brighter colors indicating higher cumulative occupancy. The dashed vertical lines delineate three modeled regions: the surrounding fluid, the superficial mucus layer, and the deeper mucus layer. Under this control condition, no blue-light input is applied to regulate bacterial motility. The simulation provides a baseline for comparing bacterial movement patterns, spatial distributions, and mucus-penetration outcomes under different light-control conditions. This video presents computational simulation results rather than experimental observations. https://2026.igem.wiki/ecust
