Title: ShellEvo: Engineering Enzymes for Green Shell Waste Valorization Description: To address the growing problem of crustacean shell waste and its associated environmental burden, we launched ShellEvo. Guided by the framework of Integrated Human Practices (iHP), we engaged six stakeholder groups along the industrial chain and incorporated their needs and suggestions into our project. ShellEvo explores a biotechnological route to convert chitosan derived from crustacean shells into chitooligosaccharides (COS) with a more defined chain-length profile. We mined protein resources from extreme environments to identify Csn-387, then used iterative protein engineering to develop a triple mutant. The mutant shifted the observed product profile from predominantly DP2–DP3 toward DP3–DP6, with DP4–DP5 as the main products, while retaining 69.84% of the parent enzyme’s catalytic activity. A key feature of ShellEvo is enzymatic hydrolysis under near-room-temperature conditions. We tested the enzyme in a 100 mL laboratory-scale fed-batch hydrolysis system at 24–26 °C to evaluate its performance under these conditions. Molecular simulations helped us develop a structural hypothesis for the shift in product profile, while stakeholder feedback informed our product positioning and production planning. Supported by the two-way synergy between stakeholder feedback and technological development, the project continuously adapts to real industrial scenarios, accommodates the demands of all parties, and steadily advances toward practical translation. It makes quantifiable and observable practical contributions to seven Sustainable Development Goals centered on SDG12 and SDG14. In the future, we will continue to advance this project, hoping to contribute to the achievement of the Sustainable Development Goals through enzyme engineering. Wiki | Official Page
