<description>Title: Kele Patch: A mRNA logic-gated RADAR sensor for cell-state specific reprogramming in keloid scars Description: Keloid scars are pathological scars which continue to grow beyond the site of the original injury and do not regress. Existing treatments include surgical removal and steroid injections, which often have high recurrence rates. We propose Kele Patch, an mRNA therapy delivered via microneedle patch straight into cells in the keloid tissue. To avoid activation in non-targeted cells, the sensor is designed to detect the transcriptional state of cells and only produce a therapeutic payload in activated keloid fibroblasts. Because of our extensive modeling of the entire pipeline from target selection to sensor design, delivery and activation, future researchers can use our platform RADAR-Scout to develop cell-state specific therapies for other diseases resulting from aberrant transcriptional programs. Kele Patch is the first synthetic biology approach to treat a widespread and overlooked disease and could easily be adapted to treat other cell-state-specific diseases, such as other fibrotic conditions.
