        <description>Title: SHUFFLE: Self-iterative High-throughput Unlocked Flexible Frequency Loci Editor Description: This is the official promotion video for the DUT-China 2026 iGEM project: SHUFFLE. Conventional microbial engineering suffers from a long-standing trade-off: random mutagenesis generates high diversity but lacks targeting specificity, while canonical base editors offer precision but only support very limited modifications. To break this bottleneck, we developed SHUFFLE, a base-shuffling editing system based on nCas9 and engineered deaminases. It enables random substitution of all four DNA letters (A/T/C/G) across multiple targeted loci within a wide genomic window, delivering both mutational diversity and targeting precision in a single pass. Coupled with EvolvePro, our active-learning Design-Build-Test-Learn pipeline, SHUFFLE can be iteratively optimized automatically. To validate its practical value, we applied SHUFFLE to engineer ethanol tolerance in E. coli. By reshuffling a library of global regulators and screening under ethanol stress, we obtained strains that tolerate ethanol and convert it into PHB, a biodegradable plastic. SHUFFLE provides a powerful, controllable foundational tool for rapid chassis cell upgrading, directed enzyme evolution, and complex trait engineering in synthetic biology.
