Title: SynAmine: Engineering Yeast for Efficient Polyamine Production Description: SynAmine is the 2026 iGEM project of SCU-BSE-China, Sichuan University: engineering the yeast Saccharomyces cerevisiae to make polyamines easier to obtain. Spermidine and spermine matter in health, agriculture and materials research, yet supply is limiting: plant sources are dilute and mixed with similar molecules, and chemical synthesis takes many steps. This fifteen-minute presentation follows two engineering routes. A multi-omics comparison of the parental strain 7D and the high-producing strain JQ43, sampled in log and stationary phase, pointed to sulfur and methionine metabolism as an underexploited upstream supply; validating the two candidate nodes separately raised spermidine by 20% (MET6, 427.03 ppm) and 12% (MET12). Because spermidine and spermine synthesis compete for a single donor, dcSAM, we then fused the donor-generating enzyme Spe2 to the spermine synthase Spe4. The forward fusion produced 214.20 mg/L spermine, 1.84 times separate expression, and a linker library yielded parts tuned for either spermine or total polyamine output. The video follows the engineering cycle behind these results, with the supporting evidence at each step: colony PCR verification, five-day fermentations and HPLC quantification. It closes with the stakeholder engagement that shaped the project, the founding of our company, and 37 new parts and composite parts. Wiki | Official Page
