<description>Title: TetraxSkin Description: How can we connect natural ingredients, synthetic biology, and DNA nanotechnology to address the needs of sensitive skin? We are JLU-NBBMS, and in this video, we share our journey toward a skincare platform designed around oxidative stress, inflammation, and practical production. Our project begins with ginseng. Using public gene-expression data and pathway analysis, we prioritized ginsenoside Compound K (CK) for experimental investigation. We then engineered Saccharomyces cerevisiae to produce CK and designed secreted enzyme modules to process polysaccharides extracted from ginseng stems and leaves. Together, these approaches explore the complementary potential of ginseng-derived ingredients while making better use of plant materials. To investigate delivery, we turned to tetrahedral framework nucleic acids (tFNA): programmable DNA structures assembled from four complementary strands. We combined sequence prediction, molecular docking, and molecular dynamics simulations to identify promising designs for CK association, then selected a candidate for experimental assembly. Alongside this work, we developed kinetic, fermentation, and skin-transport models to investigate production bottlenecks and guide future experiments. We also evaluated our formulation through cellular experiments, examining compatibility and responses associated with inflammation, oxidative stress, senescence, and skin-barrier function. These studies provide a foundation for further investigation, while our biosafety design considers containment during production and the removal of live engineered yeast before formulation. Beyond the laboratory, conversations with consumers, clinicians, researchers, and industry professionals helped shape our decisions. Our education activities created different entry points into synthetic biology for learners with diverse backgrounds and needs. Through this project, we aim to connect scientific design, experimental evidence, and community perspectives as we explore new possibilities for sensitive-skin care.
