Title: GLYCO‑TAM：Glycosylation‑engineered Tumor‑Associated Macrophage for Universal Cancer Immunosensitization Description: Despite advances in tumor immunotherapy, macrophage-driven immunosuppression and tumor immune resistance greatly limit treatment outcomes for solid tumors. Conventional immunotherapies partially suppress tumor growth yet lack universality, stable phenotypes and clinical accessibility. Our GLYCO-TAM project develops a synthetic biology strategy to remodel glycosylation of tumor-associated macrophages (TAMs). After seven DBTL cycles, MGAT5 was confirmed as the key target to construct anti-tumor macrophage phenotypes, with broad efficacy verified in liver and colorectal cancer models. We optimized an LNP-mRNA delivery system to reprogram TAMs toward the M1 phenotype and boost anti-PD-1 effects in mice. Combined with policy, safety, ethics research and bilingual science outreach, this work proposes a scalable approach to reverse tumor immunosuppression. Validated only in cell and animal models without human trials, our strategy represents a promising alternative to conventional tumor immunotherapy. Wiki | Official Page
