        <description>Title: BRO[K]EN (Building Resilient Optimized Engineered Networks) Description: While synthetic biology has produced an impressive array of engineered genetic circuits, like any engineered system, synthetic circuits break. Although several mechanisms of failure are known—mutation, selection, cellular burden, environmental fluctuations, and host interactions—there is no general framework for testing or predicting when, how, or why circuits break. To address this foundational gap, we present BROKEN: Building Resilient Optimized Engineered Networks! BROKEN employs a three-pronged approach used in mature engineering disciplines, one that will detect, predict, and help prevent circuit failure. Through principles of failure-analysis, which distinguish more mature engineering disciplines, our project aims to create synthetic biological systems that are not only functional, but predictable, dependable, and resilient — and therefore safe and sustainable.
