TU Delft: MOSAIC Designer Tutorial (2026) [English]

Most commercial gene synthesis providers will not produce highly repetitive sequences under standard pricing. Repetitive genes fail the sequence-complexity filters that vendors run before a synthesis order is accepted, so the order is either rejected outright or pushed into a bespoke, high-cost, long-lead-time service. This is not an arbitrary policy. The repetitive sequences create problems with synthesis as the DNA complex and mis- or self-anneal. For iGEM teams working with repetitive sequences like Elastin-like polypeptides, synthesis is a critical bottleneck. This creates a significant disadvantage since major iGEM sponsors like IDT and GenScript restrict synthesis of repetitive DNA. We have designed, built and validated a browser-based designer "MOSAIC Designer" that generates synthesis-ready library sequences, and that ships with our tested MOSAIC library genes. Check it out on our wiki
