Title: Cervixa: A homologous recombination-mediated biosensor for cervical cancer Description: Cervical cancer is the leading cause of cancer-related deaths among women in over 37 countries. Paradoxically, the World Health Organization estimates that 99% of these deaths are preventable by early screening. Traditional methods, such as Pap smears, carry drawbacks: Human Papillomavirus (HPV) infection, the cause of nearly all cervical cancers, may incubate for over 15 years before Pap-smear detection is feasible. We address this gap with a homologous-recombination-based biosensor that detects HPV DNA in menstrual fluid, permitting an at-home, self-administered screen free from the sociopolitical barriers of current exams. Further, our biosensor detects infection years before tumorigenic cells appear. We also present a rate-kinetic model of the homologous-recombination-mediated genetic circuit producing our readout. Because the model is built from ordinary differential equations, we can integrate it with other genetic components to form sequence-specific DNA logic gates. In summary, we present a biosensor with socioeconomic and biomedical advantages over traditional exams. Wiki | Official Page
