Title: LUMEN Description: Colorectal cancer is the third most common cancer worldwide and one of the most preventable, but colonoscopy is invasive, and many people avoid screening. Team iGEM NU Kazakhstan presents LUMEN, a non-invasive screening pathway that runs from a urine sample to the final report. DAScovery: an engineered E. coli biosensor detects N1,N12-diacetylspermine (DAS), a urinary biomarker raised in colorectal cancer. The cells turn it into green fluorescence, and a mathematical model of the system is published as the open platform “Will It Glow?”. Smartphone reader: a 3D-printed box turns an ordinary phone camera into a fluorescence reader. It uses a built-in reference to keep readings comparable and was linear from 312.5 nM to 2.5 µM in testing. ColoRisk: an AI “second pair of eyes” for colonoscopy. It outlines suspected polyps, counts each one once, and classifies them as adenoma or hyperplastic. The clinician stays in charge of every decision. On unseen data it reached 92% precision and 87% recall for detection and about 90% accuracy for classification. Also covered: our seven engineering design cycles, our human practices work with clinicians, researchers and engineers, and our “Firefly” education programme. It reached 4,721 people directly and 175,046 through social media, with 18 partner organisations, and was delivered in Kazakh, Russian and English. Wiki | Official Page
