        <description>Title: Glycoengineered TAM: Macrophage Reprogramming for Cancer Therapy Description: Despite checkpoint inhibitor approval for hepatocellular carcinoma, objective response rates remain below 20% — driven largely by M2-polarized tumor-associated macrophages that dominate the tumor microenvironment.To address this,our team has developed GlycoHepaMac, a synthetic glycoengineering platform targeting a key glycosylation enzyme in tumor-associated macrophages (TAMs).GlycoHepaMac focuses on regulating glycosylation mediated by this TAM-secreted enzyme, a critical factor driving that pushes macrophages toward a tumor-promoting state and suppresses anti-cancer immune activity.Through glycoengineering inhibition of this enzyme, we redirect TAMs to the anti-tumor M1 phenotype and relieve local immune suppression, thereby restoring the therapeutic potency of immunotherapy.For better clinical translation, we developed liver TAM-specific delivery carriers to precisely modulate this glycosylation signaling.This glycobiology-based strategy extends beyond liver cancer to other macrophage-rich solid tumors, opening an innovative route for broader tumor immunomodulation.
