        <description>Title: Dual-Response EcN: An Adjunct Therapy for Pseudomonas Infections in cystic fibrosis Patients Description: Patients with cystic fibrosis frequently experience recurrent drug-resistant infections due to chronic biofilm infections caused by Pseudomonas aeruginosa in the lungs, and there is a lack of effective targeted clinical treatments. We designed an EcN strain carrying a coupled Fur and RhlR sensing circuit to enable dual-specific recognition. The Fur system is only de-repressed in iron-deprived mucus at the site of infection, thereby enabling spatial localization; meanwhile, RhlR detects the AHL quorum sensing signals released by Pseudomonas aeruginosa. Only when both a low-iron environment and high AHL levels are simultaneously detected is the expression of alginate lyase and lysis E triggered. Lysis E cleaves E. coli, releasing alginate lyase that degrades the biofilm matrix of Pseudomonas aeruginosa, thereby enhancing the efficacy of antibiotics. This synthetic biology design integrates environmental and pathogen sensing functions, establishing a strategy to achieve targeted localization and in situ clearance of biofilms in chronic respiratory infections.
