<description>Title: SUNRISE: A New Dawn in the Fight Against Antimicrobial Resistance Description: Antimicrobial resistance (AMR) is one of the most urgent public health challenges facing modern medicine. As resistance spreads, effective antibiotic options continue to narrow, leaving patients with fewer treatment options. To overcome this challenge, we propose SUNRISE (Safety-enhanced UNconventional Resensitization through Injection and Selective Elimination), a system designed to restore susceptibility to existing antibiotics. This system combines a Cargo Protein Injection (CPI) module and a Targeted Protein Degradation (TPD) module. The protein-based Injector introduces the Adaptor directly into the target bacterium. Once inside the cell, the Adaptor binds to the antibiotic resistance protein through its Binder and recruits the endogenous ClpXP protease complex for degradation. Thus, this system aims to reduce resistance protein levels and restore susceptibility to existing antibiotics. By independently redesigning the Adaptor's Binder and Injector's tail fiber, SUNRISE can be rapidly adapted to diverse resistance proteins and bacteria, bringing a new dawn to the fight against AMR.
