<description>Title: PB-Shock Description: Our project aims at developing a living, locally active oral biotherapeutic platform for treating colorectal cancer. Rather than infusing cytotoxic agents throughout the entire body, our system leverages an engineered, food-grade probiotic strain Lactobacillus casei ATCC 393 to produce and secrete the human cytokine IL-37. By modifying the bacterial surface with a cadherin-targeting peptide, it specifically targets colorectal cancer cells and intended to promote preferential bacterial localisation and retention at tumour sites, enabling local IL-37 secretion. We hypothesise that this local delivery could achieve concentrations sufficient to induce antitumour effects while keeping exposure in healthy tissues below an effective level. Through continuous engagement with stakeholders, we determined how our project serves the public and evaluated the ethical, safety, and societal implications of our biotherapeutic platform. Beyond that, we designed innovative educational and outreach activities to open a two-way dialogue with new communities regarding synthetic biology and public health.
