<description>Title: SCOPE - Sustainable Cellulose: Optimizing Products for Everyone Description: Our project rests on two pillars: sustainability for the planet and accessibility in the lab. Bacterial cellulose (BC) is prized for its strength, purity and biocompatibility; environmental economists we consulted identified BC-based beauty masks as a near immediate commercial application. Life cycle analysis against four substrates found BC used the least water (6.3 mL per functional unit) but had higher carbon impacts. Upscaling BC therefore rests on yield per unit energy and feedstock - allowing us to set engineering goals. Native Komagataeibacter strains grow slowly and mutate into non-producers, so we pursued E. coli recombinant synthesis: overexpressing phosphoglucomutase and UDP-glucose pyrophosphorylase across constitutive, inducible and cold-inducible promoters to relieve the precursor bottleneck, and modeling alanine substitutions at BcsA’s Glu370–Arg583 salt bridge to predict a c-di-GMP-independent synthase. In parallel we built HALO, a wheelchair-mounted modular lab organizer co-designed with wheelchair-using scientists, removing a physical barrier to participation in synthetic biology.
