<description>Title: Computationally Optimized Space Monitoring and Omni-Sensing System Description: Long-duration spaceflight poses significant challenges to astronaut health, including accelerated bone and muscle loss, weakened immune function, and heighted physiological stress. Monitoring these responses is essential for maintaining crew health during extended missions. Here, we present a biosensing platform starting from computational design of the synthetic binders all the way to measuring their performance against key biomarkers. Using RFdiffusion 3 and BindCraft, we designed de novo binders targeting osteocalcin, a marker of bone turnover, and PD-L1, a protein involved in immune regulation. Candidate binders were prioritized using AI-based predictions and evaluated through AMBER molecular dynamics to teach our Model what features contribute most to binding affinity, leading to better design. Selected candidates were expressed in Escherichia coli, purified, and assessed experimentally for binding activity. This integrated computational and experimental workflow supports the development of a compact biosensor for space capable of monitoring astronaut physiology with limited laboratory infrastructure.
