<description>Live-cell confocal time-lapse of Saccharomyces cerevisiae expressing GCaMP6f (piGEM005), well C5, no added calcium. Recorded on 30 September 2026. Cadmium chloride was added to a final concentration of 90.9 µM approximately three minutes before this recording began. Nikon Ti2 with AX confocal scanner, S Plan Fluor ELWD 40x objective (NA 0.6), 488 nm excitation at 1.1% power, 499-551 nm emission, detector gain 44.7, pinhole 25.6 µm, 512 x 512. 61 frames at 5 s intervals, 300 s total. Nikon Denoise.ai was active during acquisition. Green fluorescence does not change over the recording: whole-field mean 157.97 at frame 1 and 157.51 at frame 61, a drift of 0.3%. The cadmium-triggered calcium transient in yeast peaks within about 20 s and resolves by 50-60 s, so this recording began after that window had closed. Credits: iGEM Saarland 2026 (OWLET, team 6311).
