Within this field, Clostridium butyricum has attracted attention as a butyrate-producing probiotic with anti-inflammatory, barrier-supporting, and microbiota-modulating properties. Current evidence suggests that it may alleviate gut-related disorders, with the strongest support in IBS-D and experimental colitis/IBD models, where it improves symptoms, reduces inflammation, and strengthens epithelial barrier function. Since C. butyricum is still a non-model organism with limited genetic tools, engineering remains challenging due to DNA delivery, gene-expression control, and genome-editing bottlenecks. In addition, as a live biotherapeutic, it must be made genetically stable, safe, and reliable in the gut environment. Therefore, we are building a toolkit for C. butyricum to support deeper study and future therapeutic development. By systematically characterising a range of promoters in this chassis and experimenting with existing promoters and mining the genomes of close relatives, our ultimate goal is to find a set of promoters that can be utilised therapeutically in the future to improve gut health.
