Title: Engineering a Two-Strain System for Naphthalene Degradation and Salicylic Acid-Based Biosensing Description: Polycyclic aromatic hydrocarbons (PAHs) are widespread, persistent pollutants; global emissions of 16 priority PAHs reached higher recent years. Naphthalene, a frequently detected PAH in urban soils and its Group 2B IARC classified. Conventional detection (HPLC/GC) is costly and laboratory-bound. HUBU- Wuhan developed a two-strain E. coli system: a degrader strain carrying the nah cluster from Pseudomonas putida G7 converts naphthalene to salicylic acid, which a sensor strain detects via a bidirectional NahR–Psal/Pr-driven eGFP reporter, i.e. NahR-salicylic acid complex activates Psal-driven eGFP. Through multi-layer engineering—promoter optimization (100 to 20 μM), NahR saturation mutagenesis (20 to 0.2 μM), and ARTP evolution (0.2 to 0.05 μM), achieved lowest detection limit of 0.05 μM, the lowest reported to date. Promoter replacement, and Fe²⁺ tuning lead to near-complete naphthalene degradation within 4 h at low concentrations (20–100 μM). Integrated portable device with mobile App, enables on-site PAH screening, leading to future field deployment. Guided by the RRI framework, our Human Practices work with regulators, enterprises, and the public deeply reshaped our project design. Our video presentation showcases our journey of translating synthetic biology into a practical tool for environmental remediation. Wiki | Official Page
