<description>Title: GENELOCK: a new antibiotic mechanism based on programmable DNA sequestration Description: Antimicrobial resistance is an emerging threat to public health, motivating the search for novel bacterial inhibition mechanisms. Since recent work demonstrated that synthetic polymers can sequester bacterial DNA through phase separation, inhibiting transcription and inducing cell death, we investigated genetically encodable protein condensates as a programmable alternative. We computationally designed a modular condensate-forming protein with high-affinity DNA binding domain, intrinsically disordered region to drive condensation, and oligomerization domain to increase valency. To determine whether DNA sequestration interfered with gene expression, we used an in vivo ParB–parS reporter to assess localized DNA sequestration and transcriptional repression. Finally, we quantified bacterial growth and viability using optical density measurements and colony-forming unit assays to evaluate antimicrobial activity and proved protein expression via GFP-tagged protein. Together, these experiments investigated whether programmable protein condensates could compact bacterial DNA, inhibit transcription, and provide a novel antimicrobial strategy.
