<description>Title: Omni-Patch Description: Chronic wounds frequently become trapped in persistent inflammation, disrupting tissue repair. Omni-Patch addresses this challenge through a cell-free hydrogel dressing combining a de novo-designed peptide with the tissue repair factor FGF2. Our de novo computational pipeline integrates RFdiffusion, LigandMPNN and molecular dynamics simulations to generate and screen peptide candidates targeting the MD-2/LPS interface of the TLR4 signaling complex. Experimental screening identified αTi-14 as a promising lead that reduced cell-associated fluorescent LPS in human macrophage-like cells. Through iterative Design-Build-Test-Learn cycles, we refined cellular assays to investigate its anti-inflammatory potential; functional confirmation remains ongoing. In parallel, we explored recombinant peptide production and alginate delivery, with preliminary release studies using FGF2 and peptide variants. Mathematical modeling connected molecular design with delivery kinetics and downstream biological responses. Guided by stakeholder engagement and educational initiatives, these efforts establish a foundation for developing Omni-Patch to locally modulate excessive inflammation while supporting tissue repair in wounds.
