Title: AETh3R: A PROTAC-Based Strategy for Targeted Protein Degradation in NSCLC Description: Non-Small Cell Lung Cancer (NSCLC) is the most common form of lung cancer, with treatment resistance and disease recurrence remaining major clinical challenges. iGEM Thessaloniki 2026 is developing AETh3R, a novel PROTAC-based therapeutic approach designed to target key oncogenic proteins, including MDM2 and KRAS. AETh3R harnesses the cell’s own proteasomal machinery to selectively degrade these proteins, disrupting cancer cell survival pathways and promoting apoptosis. By combining a target-binding domain, flexible linker, and E3 ligase-recruiting ligand, our heterobifunctional molecules aim to eliminate oncogenic drivers at the protein level. Through targeted delivery and precise protein degradation, AETh3R offers a promising strategy to overcome limitations of conventional therapies and tackle treatment resistance in NSCLC. By bringing together synthetic biology, targeted protein degradation, and translational thinking, we envision a new direction for more precise cancer treatment. Wiki | Official Page
