There is no transcription factor in LGG that binds DEHP. So the circuit reads the consequence instead. As DEHP accumulates in the acyl region, chain order rises, area per lipid falls, and the bilayer thickens and stiffens. DesK senses that physical change, autophosphorylates at its conserved histidine, and hands the phosphoryl group to DesR. A membrane state becomes gene expression. 00:00 Chain order rises with the DEHP load 00:06 DesK senses rigidity, not DEHP 00:12 DesK~P to DesR~P, and the operator Chapter 4 of 7 from the FSU iGEM 2026 project Lilieum. DesK/DesR is the sensor we are building toward. Our current constructs use inducible promoters as a testbed. https://2026.igem.wiki/fsu
