<description>Title: Temperature-Controlled Smart Odor-Degradation System Based on Engineered Microorganisms Description: Odor pollution is a prevalent problem in modern daily life. Foot odor, pet waste and garbage piles continuously release volatile short chain fatty acids. Isovaleric acid is the core odor causing substance responsible for sour and rancid smells. Trace amounts of isovaleric acid can generate strong unpleasant odors and greatly reduce people’s living comfort. In our project, we constructed multiple basic and composite genetic parts. We selected Escherichia coli as the expression host and finished the construction of recombinant plasmids. The complete degradation pathway of isovaleric acid requires four key proteins. They are FadL, FadD, AidB and MccAB. We designed the degradation module and built three recombinant plasmids. The expression of partial genes may lead to insufficient substrate activation or accumulation of toxic intermediates. Only the co expression of all four genes can achieve efficient and complete degradation. Meanwhile, we introduced the pTRIP temperature dependent quorum sensing dual regulation system to avoid ecological risks caused by engineered strain escape. We designed a lethal module and constructed three recombinant plasmids. The suicide pathway remains off under fermentation conditions at 37 °C. If strains escape into low temperature natural environments, the lysis and lethal mechanism will be triggered with the coordination of AHL signals to guarantee biosafety. Our project adopted synthetic biology strategies to assemble the isovaleric acid degradation pathway and temperature controlled suicide module into Escherichia coli, constructing safe and efficient odor removing engineered bacteria.
