Title: DigiActivate: Chemically Induced, Tunable Gene Activation Description: Cell fate is governed by fluctuations in gene expression over time, yet CRISPR transcriptional tools are binary, holding a gene ON or OFF at a fixed level over a fixed period. Engineering that timing and magnitude is what therapies for haploinsufficiency or T cell exhaustion require. DigiActivate achieves this by fusing an array of inducer gated switches onto dCas9, where each switch controls the recruitment of a different transcriptional activator. We tested multiple levels of activation on a two switch array through flow cytometry, where we observed distinct 1.3, 1.7 and 1.9 fold increases in eGFP mean fluorescence intensity corresponding to inducer additions of B, A and A+B respectively. Finally, we used RFdiffusion to design compact de novo binders replacing the natural, bulky E3 ligase domains. The result is a modular part collection in which the inducer or the effector can be changed by swapping a single part. Wiki | Official Page
