<description>Title: The Great Wall: Engineering Light-Programmable Living Microrobots for Precision Mucosal Vaccination in Aquaculture Description: Evolved from last year’s Wukong project, The Great Wall is an upgraded industry-oriented aquaculture vaccine solution from East China University of Science and Technology, addressing the fish mucosal barrier bottleneck that causes over US$5 billion in annual global disease losses. The prior project had two core limitations: its transcription-level optogenetic control suffered minute-scale response lags mismatched with bacterial motility, and its first-generation screw immersion system had poor steady-state performance with no vaccine fluid circulation. Moving away from the original transcriptional light-controlled expression strategy, we adopted protein-level allosteric engineering as our core design paradigm — a more direct, kinetics-aligned approach tailored to bacterial motion regulation. By fusing the AsLOV2 photosensitive domain with chemotaxis protein CheZ, we built a second-scale light-responsive allosteric system that fundamentally resolves the speed mismatch. Through systematic DBTL engineering cycles and AI-aided linker optimization, we achieved far more precise and stable motility regulation than the previous generation. We also established a complete in vitro quantitative toolchain including ABM simulation, motion-tracking algorithm and biomimetic microfluidic chip, filling the gap of phenotype-only observation in the prior work. For industrial deployment, we upgraded the screw system into an intelligent steady-state vaccine workstation with integrated fluid circulation, multi-parameter monitoring and model-optimized light module, translating isolated component breakthroughs into a systematic immunization solution. Guided by the iHP framework and refined through on-site industrial investigations, the project aims to fill the fry immunization vacuum, reduce antibiotic dependence, and deliver a mature, deployable engineering solution for blue food security.
